"Investigate the trail" is a mission objective in Sequence 7 / Memory 03: Confrontation

Walkthrough

Activate Eagle Vision. You will see small clouds of gold highlight on the ground. This is your trail to start following. head through the nearby archway and start going to the south past the Alarm Bell. You can either sneak past the Soldiers or take them out quietly. Just continue to the Southeast after that.

When you leave the restricted area, the trail will have a break. Look to the South. You will see it resume going upwards. Head toward the church. This will trigger a cutscene.
