"Lose Madrazo's men" is an objective in the story mission Marriage Counseling in Grand Theft Auto V.

After that brief trip toward home, things did not work out for the better. That lady was apparently Mrs. Madrazo and she just sent a hit squad after you.
You have the choice of either gunning down the people in the car (killing 3 of them counts toward the Gold Medal), or losing them with some high speed tricks. A little of both is recommended. That or stop and gun the people down from inside the car. After that, just head back to Michael's Mansion
